      目的:為建立脫落酸的熒光檢測方法. 方法:通過重疊PCR技術,在水稻脫落酸受體PYL2蛋白的C-端融合一種新型黃綠色熒光蛋白mNeonGreen,并對融合蛋白進行原核表達和純化. 結果:獲得到了高純度的融合蛋白PYL2-mNeonGreen,加入脫落酸后,該融合蛋白黃綠色熒光強度逐步降低,表明融合蛋白PYL2-mNeonGreen能特異性識別脫落酸. 結論:該研究為后續脫落酸的原位檢測奠定了基礎.
英文摘要:
      Objective: To establish a fluorescence detection method for abscisic acid. Method: A novel yellow-green fluorescent protein mNeonGreen was fused to the C-terminus of the abscisic acid receptor PYL2 from rice by overlap PCR, and the fusion protein was prokaryotic expressed and purified. Results: The fusion protein PYL2-mNeonGreen with high purity was obtained. With the addition of abscisic acid, the yellow-green fluorescence intensity of the fusion protein was gradually decreased, indicating that the fusion protein PYL2-mNeonGreen can specifically recognize abscisic acid. Conclusion: This study laid the foundation for the in situ detection of abscisic acid.
